Termination tasks
Center the Epi-illumination lamp. (It should be executed each time
you replace the Epi-illumination lamp.)
Adjust the diopter and the interpupillary distance.
A TV camera or dust-proof cap is in place on each of the ports on the
microscope.
An objective or dust-proof cap is in place on each of the sockets of
the nosepiece of the microscope.
The fiber is securely fixed.
The Epi-illumination lamphouse is attached.
The cassette holder is attached. (Its lid is also closed.)
Emit the laser.
Turn on other components. (Microscope, Epi-illumination lamp,
shutter control unit, camera, etc.)
Open the eyepiece shutter.
Change the optical path.
Select the excitation method.
Open the Epi-fl attachment shutter.
Bring the specimen into focus.
Center the field diaphragm and adjust its size.
Adjust the brightness with the ND filter.
Close the Epi-fl attachment shutter.
Change the excitation method settings. (Place the filter cube, which
has settings of "G excitation", "Dichroic mirror for UV", and "No
barrier filter", in the optical path.)
Set the aperture diaphragm to its minimum setting.
Adjust the field diaphragm opening to the minimum required for
viewing.
Close the Epi-fl attachment shutter.
Close the Epi-fl attachment shutter.
Close the eyepiece shutter.
Change the optical path.
Select the excitation method.
Open the laser shutter.
Collimate the laser beam.
Adjust the laser position to provide TIRF illumination.
(When magnification is changed, repeat above steps from a step
"Collimate the laser beam.")
Turn off the laser.
Turn off the Epi-illumination lamp.
Turn off other components.
